In vitro polymerization of flagellin excreted by a short-flagellum Salmonella typhimurium mutant.

چکیده
The culture medium of a short-flagellum mutant of Salmonella typhimurium contained a large amount of mutant flagellin and a small amount of strong inhibitors of flagellin polymerization. After being freed of the inhibitors, the mutant flagellin could be polymerized in vitro, although under nonphysiological conditions.

Excretion of flagellin by a short-flagella mutant of Salmonella typhimurium.
A nonmotile mutant of Salmonella typhimurium, SJW1254, has very short flagella (less than 0.1 micron long) due to a mutation in the structural gene of flagellin (H2). When ammonium sulfate was added to the culture medium of SJW1254 grown to the late-log phase, a large amount of protein precipitated. Gel electrophoresis and immunodiffusion showed that more than 90% (wt/wt) of the precipitated pr...
متن کاملIn vitro reconstitution of flagellar filaments onto hooks of filamentless mutants of Salmonella typhimurium by addition of hook-associated proteins.
An in vitro system for reconstituting flagellar filaments onto hooks of filamentless mutants of Salmonella typhimurium was used to investigate the role played in filament formation by the three hook-associated proteins (HAPs, products of the flaW, flaU, and flaV genes). These proteins--FlaW, FlaU, and FlaV--are believed to be assembled in this order at the distal end of the hook. When the recip...

ABSTRACT Background and Objective: In this study, we compared the susceptibility of Salmonella typhimurium phoP mutant and its parent to stress conditions that the pathogen may encounter in a host. Methods: For this purpose, we used the phoP deletion mutant constructed in our previous study. In order t...
متن کاملSalmonella flagellin is not a dominant protective antigen in oral immunization with attenuated live vaccine strains.
We found that oral immunization with flagellum-defective mutant strains of Salmonella enterica serovar Typhimurium with the ClpXP-deficient background protected mice against oral challenge with the virulent strain. These data indicate that Salmonella flagellin is not a dominant protective antigen in oral immunization with attenuated live vaccine strains.
متن کاملEnhancement of Host Immune Responses by Oral Vaccination to Salmonella enterica serovar Typhimurium Harboring Both FliC and FljB Flagella
Flagellin, the structural component of the flagellar filament in various motile bacteria, can contribute to the activation of NF-κB and proinflammatory cytokine expression during the innate immune response in host cells. Thus, flagellin proteins represent a particularly attractive target for the development of vaccine candidates. In this study, we investigated the immune response by increasing ...
